The balance between T helper type 1 (T H1) and other T H cells is crucial for antiviral and anti-tumour responses, yet its regulation is poorly understood.
We identified regulators modulating T helper cell differentiation using a unique T H1-T H2 cell dichotomous culture system validated through in vitro and in vivo CRISPR screens.
RAMP3, a receptor component, has a critical role in T H1 cell fate determination, promoting their differentiation while restricting T H2 cells through CGRP signaling.
Post-viral infection, a neuroimmune circuit involving CGRP from neurons and RAMP3 on T cells enhances anti-viral responses, critical for controlling acute infections.
