Understanding the developmental dynamics of layer 5 extratelencephalic neurons is critical, as they play a key role in fine motor skills and are vulnerable to spinal cord injury and amyotrophic lateral sclerosis.
We identify subtype-specific developmental dynamics of ET neurons, revealing that ET prox neurons emerge through the pruning of ET dist axons or de novo from common progenitors.
Our findings highlight the functional transcriptional programs that govern the diversity of ET neurons and present opportunities to manipulate their connectivity via gene regulatory networks.
By utilizing single-nucleus sequencing, we outline the transcriptional programs that can be leveraged to direct motor corticofugal pathways, enhancing therapeutic interventions.
