Orforglipron, a once-daily oral GLP-1 agonist, reduced body weight by up to 20% in a clinical trial in which one in five participants lost 20% or more over 72 weeks. The randomized study enrolled 3,127 adults with obesity (BMI ≥30) without diabetes across multiple countries and compared different pill strengths to placebo. Orforglipron acts by lowering blood sugar, slowing gastric emptying and reducing appetite. The weight loss observed was less pronounced than with tirzepatide injections, but the oral formulation offers easier storage, distribution, administration and potential cost advantages. Orforglipron is not yet approved by the FDA or other regulators.
