A man in his 50s and a woman in her 40s were treated for non life-threatening injuries at Letterkenny University Hospital, while an elderly woman was taken for serious injuries.
Gardaí are appealing for witnesses who were on the N56 between 8.30am and 9am to provide any camera footage to assist with the investigation.
The crash drew attention following a separate fatal incident earlier in the day where a 16-year-old lost his life after a car crash in Inishowen.
Investigations are ongoing into the collision, and the N56 road remains closed as authorities work to gather evidence and assess the situation.
