The central conclusion of Chen et al. regarding the role of KLHL22 in controlling DEPDC5 stability and mTORC1 activation is undermined by our findings.
Our investigation using validated antibodies reveals that DEPDC5 stability is not influenced by amino acids, proteasome activity, or loss of KLHL22 function.
These results suggest that the mechanisms linking nutrient availability to mTORC1 activation are more complex than previously proposed by Chen et al.
