Chromatin formation requires a steady supply of nucleotides and histones. PRPS enzymes catalyze the first step in nucleotide synthesis and additionally participate in preparing histones for chromatin assembly. Loss of PRPS1 or PRPSAP1 severely disrupts histone maturation, indicating a role separate from nucleotide production. PRPS involvement in early histone maturation effectively links nucleotide biosynthesis with histone production. This linkage synchronizes DNA replication and chromatin assembly so that both processes proceed at matched rates, promoting accurate chromatin construction during DNA synthesis.
