"Progesterone is one of two key female sex hormones, the other being estradiol, better known as oestrogen. Typically found in pill form, progesterone can be taken either orally or rectally, known as boofing. While it tends to promote breast growth in cisgender women, there have been few studies on its safety and effectiveness for transgender women. The lack of reliable evidence means progesterone supplements are rarely prescribed to trans people."
"Presenting the results at the European Professional Association for Transgender Health's (EPATH) annual conference in Hamburg, Germany, on Friday (5 September), endocrinologist Koen Dreijerink said researchers found that, not only was the medication "safe and effective" but it also dramatically increased breast growth. "We're now able to prescribe it, in a trial setting, for those who have been taking [oestrogen] for at least a year," he said. "We hope our findings lead to better hormone treatments for transgender individuals.""
A medical trial at Amsterdam University Medical Center enrolled 90 participants who were taking feminising hormone replacement therapy. Progesterone was added to existing estradiol treatment and administered in pill form or via rectal routes. Breast size and volume were measured with 3D-scanning techniques, showing an average increase of about 30 percent. The greatest increases occurred in participants who also increased estradiol dosage. Reported side effects included short-lasting tiredness, hypersensitivity of breasts and nipples, and mood swings. Progesterone supplements have been rarely prescribed previously due to limited evidence. Long-term safety and side-effect profiles of prolonged progesterone use remain to be clarified.
